Mahomes broke Len Dawson’s franchise record of 30 touchdown passes in one season when he connected with Tyreek Hill on a 14-yard scoring play in the second quarter against the Arizona Cardinals.
It was the second time the pair had combined for a touchdown in the game, with Hill having already caught a 37-yarder to put Kansas City 7-0 ahead.
Mahomes, who is in his second year in the league, was making just his 11th NFL start on Sunday.
He set a new league record for touchdown passes in the first three games of a season this year, including six against the Pittsburgh Steelers in Week 2.
The NFL’s single-season touchdown record sits at 55, set by Peyton Manning in the 2013 season with the Denver Broncos.
